제목: Dimorphism and Enzyme Activity of Saccharomycopsis fibuligera Dependent on pH changes during Fermentation



내용 :

Saccharomycopsis fibuligera is dimorphic yeast frequently found in Korean fermenting starter nuruk. This Yeast strain has high starch degradation activity indicating its main role of saccharification in makgeolli fermentation. During fermentation of Korean traditional rice wine, pH change from the neutral to acidic has been known. However, little is known about the impact of the pH change on the phenotypic characteristics of S. fibuligera. Scanning electron microscopy reveals that pH change induces the transmission of yeast cell formation vs. hyphal growth in S. fibuligera KHP12 (a single genome, 19Mb) and its interspecies hybrid KJJ81(hybrid genome, 38Mb). In addition, the enzyme activities of starch and cellulose degradation have been decreased by changing pH. Based on these phenotypes, we performed RNA-Seq analysis and found that the genes of 62% and 74% in each KHP12 and KJJ81 were differentially expressed from pH7 to pH3. By performing soft clustering analysis and transcriptomic analysis, we identified the sets of genes related to dimorphism and carbohydrate metabolism that are differentially expressed. Until now on, we found that the genes associated in Rim101 and cAMP-PKA signaling cascades may regulate the dimorphism and enzyme production in S. fibuligera.
